Place a shop towel near the nipple end of the air bleed screw and hit the primer button until you see all diesel and no air coming out.....usually not much more than 6 to 8 pumps if you primed the filter with diesel fuel supplement right the first time.

A self priming pump is like any other centrifugal pump, but it is designed with an external casing that “floods” the inner pump or volute. The casing is filled with liquid [usually the same as the liquid to be pumped] and the pump is ready to go.
Put one of those in the fuel line down by the fuel pump, pointed in the correct direction. You will find that will prime the line up to the fuel pump. Once you do that then you can use the puffs of vacuum on the pump to check whether the pump is actually working.

You don't need to prime the fuel pump, mechanical pumps are self priming. Just dump a little gas (like a tablespoon) straight into the carb and fire the engine up. It might take one or two attempts before the engine will stay running on its own, but there is no real need to prime IMO.
